ABSTRACT

CBDA synthase was purified from the homogenate of leaf buds of CBDA strain by using DEAEcellulose, phenyl Sepharose CL-4B and hydroxyapatite, successively. CBDA synthase of Mexican strain on DEAE-cellulose, phenyl Sepharose CL-4B, and hydroxyapatite, successively. cannabidiolic acid (CBDA) synthase was confirmed to be homogeneity having 75 kD by SDS-PAGE. The enzyme does not require coenzyme, hydrogen peroxide, molecular oxygen and any metal ion for cyclization of geraniol moiety resulting in not an oxygenase or a peroxidase. Tetrahydrocannabinolic acid (THCA) synthase and cannabichromenic acid (CBCA) synthase were purified from the homogenate of leaf buds of Mexican strain and young leaf buds of CBDA strain respectively as the same manner with that of CBDA synthase. Purified THCA synthase was interestingly resemble to that of CBDA synthase only except the catalyzing ability for CBGA to THCA. It became evident that CBCA synthase catalyzes CBGA to two CBCA epimers as a ratio of 5:1.
