ABSTRACT Acetaminophen (APAP) overdose is a leading cause of liver failure. and probably also acute renal failure. This study aims to analyse the acute and sub-chronic damage of liver and kidney function after a single toxic oral dose of APAP in Wistar rats. The control group was treated with saline solution and the experimental groups were treated with 3 g/kg of APAP. We observed in the acute and sub-chronic damage groups that APAP increased plasma AST, ALT, MPO, MDA and nitrite levels and, decreased GSH in the liver and kidney, without change in urea and creatinine. Liver tissue showed degenerative changes in the hepatocytes and cells lining the blood sinusoids, necrosis, hemorrhage, inflammatory cell infiltration, edema and congestion; and in the kidneys, congestion, turbid swollen tubulitis with accumulation of proteinaceous material within the tubules, and inflammatory cell infiltration were observed. We conclude that APAP produces greater hepatic and renal damage after 7 days of ingestion compared to damage produced after 24 hours.
