ABSTRACT
 
The mechanism underlying release of neurotransmitter from nerve terminals is, at the present writing, not fully resolved. In particular, it is not known how the process of release initiates. In other words, how with the release system and all its components “ready to go” an abrupt signal launches the execution of the release process. In this review we provide data suggesting that it is the presynaptic autoreceptor which controls initiation of release.
