ABSTRACT
Barely (Hordeum vulgare L.) is a potential new source of genes for wheat (Triticum aestivum L.) improvement. We previously reported that barley chromosome 5H has genes for early-heading which function in the wheat genome. To introduce the barley genes for early-heading on chromosome 5H into wheat, we are producing wheat-barley 5H chromosome recombinant lines using 5B nullisomy and molecular markers. For the effective screening of recombinants involving 5H, we developed 18 barley-allele specific PCR markers (amplicons) which can be distinguished from their wheat counterparts without restriction enzyme digestions for chromosome 5H. Among the progeny of the line with nullisomic-5B trisomic-5A monosomic 5H (19II+5AIII+5HI) lacking the Ph1 gene on chromosome 5B, we were able to detect several plants having putative wheat-barley recombinant chromosomes by using 5H allele-specific amplicon makers.
