ABSTRACT

Two simple, rapid and accurate spectrophotometric methods are presented for the determination of amantadine in pure form and in pharmaceutical preparations. In the first method, three acid dyes have been utilized for the spectrophotometric determination of amantadine. The dyes used were bromocresol green (BCG), bromophenol blue (BPB) and bromocresol purple (BCP). In chloroform; they formed stable yellow ion-pair complexes with choloroformic solution of amantadine which absorbed at 408 nm for both BPB and BCP and at 416 nm for BCG. The second method is based on the reaction of amantadine in dimethylformamide with ascorbic acid at 100 ºC for 15 min. Afterwards the absorbance of the reaction product is measured at 420 nm. The two procedures were applied successfully to commercial capsules, the results obtained were in good agreement with the obtained by the official method.
