ABSTRACT

The Grb7 family of adapter proteins is presently formed by Grb7, Grb10, Grb14. They are related to the Caenorhabditis elegans Mig10 protein which is involved in the regulation of embryonic cell migration. The Grb7/Grb10/Grb14 proteins are downstream signalling molecules of tyrosine kinase receptors, as they bind to the phosphotyrosine residues of activated receptors via their SH2 domain. Grb7 appears to be involved in the regulation of cell migration, and it is found co-amplified with ErbB2./Neu in adenocarcinoma cells, suggesting a role in extramucosa tumour invasion. Grb10 on the other hand appears to regulate, among other functions, cell proliferation and other signaling events mediated by the insulin receptor. This review will summarise the major features concerning the physiological roles of these adapter proteins, and the possible implication of the Ca2+ signal in the modulation of their functions.
