ABSTRACT
 
In Okinawa, located in southwest island of Japan and in subtropical area, the incidence of lung cancer cases showing squamous cell carcinoma in histology is higher than that of mainland in Japan. Especially the proportion of well differentiated squamous cell carcinoma was quite higher than that of mainland. We found the correlation between squamous cell carcinoma of the lung in Okinawa and HPV infection by means of PCR and in situ hybridisation. Among the HPV detected cases, most prevalent types were high risk type HPV such as HPV16 and HPV18, however the proportion of low risk types, HPV6 and HPV11 comprised small proportion. We postulated that HPV induced morphological changes (i.e. from adenocarcinoma to squamous cell carcinoma). In order to examine the relationship between morphological change and HPV infection, HPV 16 and 18 were transfected into cultured adenocarcinoma cell line and subsequently they were transplanted to SCID mouse. HPV transfected adenocarcinoma showed squamous differentiation by histological analysis. In the lesion, high molecular weight keratin and involucrin expression were demonstrated by Western blotting and immunohistochemistry. The growth rate of HPV transfected cells was decreased and cell cycle analysis showed increased G0-G1 population. HPV transfected cells showing marked squamous differentiation exhibit high expression level of E6, E7 and E2. In order to identify responsive gene for squamous differentiation, we transfected single HPV gene (E6, E7 or E2) into adenocarcinoma cells. The expression level of cell cycle related gene and apoptosis related genes were decreased and cell adhesion molecules were increased in HPV single gene transfected cells. Although E6 expressing strain showed higher expression of squamous differentiation related genes than other strains, the morphological change and the expression level of squamous differentiation related genes were not so distinct as multiple HPV gene expressing cells. Our data suggests that simultaneous HPV genes and cellular genes interaction is required for morphological change.
