ABSTRACT Lanthanide fluorescence chelates have been successfully developed as the fluorescence labels for the highly sensitive bioassays in recent 20 years. The applications of lanthanide fluorescence labels for time-resolved fluoroimmunoassay (TR-FIA), DNA hybridization assay, cell activity assay, and fluorescence imaging microscopy have been widely investigated. The advantage of time-resolved fluorescence measurement using lanthanide fluorescence chelate as the label is that the method makes easy distinction of specific fluorescence signal of the long-lived label from short-lived background signal present in most biological samples, and also obviates problems associated with scattering light of the optical components, whereas these problems can not be solved easily by the general fluorescence measurement method. Recently, the developments of time-resolved fluorometry for bioassays using lanthanide fluorescence labels have been directed mainly to the following topics, (1) synthesis and application of new lanthanide fluorescent labels, (2) developments of new principle and method and (3) combination with other analytical techniques. In this paper, the developments of lanthanide fluorescence chelates and their applications in bioassays are reviewed based on our works and recent publications.
